MODIFIED OLIGO ORDER FORM INSTRUCTIONS

This page was developed to instruct users how to place oligo orders using our MODIFIED OLIGO ORDER FORM which can be accessed by clicking the green "Order Modified Oligos" button on the W.M. Keck Oligo Synthesis Home Page.

This page includes screen captures showing examples of how to fill out this form, but keep in mind that the examples here will vary slightly from what you see depending on your browser and operating system. The site accepts orders for all modified oligos, including chimeric DNA/RNA. Please place orders for normal DNA, RNA, and 2'-O-methyl RNA via the Normal Oligo Order Form (Click the "Order Normal DNA or RNA" button on the ordering page.

Upon entry to the site you will be prompted to enter your password. Do so in the box provided and click "Order Modified Oligos". If you need a password, please see our Registration Page. If your password is valid you will gain access to the MODIFIED OLIGO ORDER FORM. Check to see if the information (your password, your name, your PI's name, etc...) is correct. If not, please contact us.

At this point, you will need to enter your billing information and a valid E-mail address where we can reach you. See the screen capture below:

Enter a current Yale PTAEO, or if you're a non-Yale researcher, a Purchase Order number. Then enter a valid E-mail address.

You will then fill out the "Oligo Name / Sequence / Modification Information" fields for each oligo. Please use a unique Oligo Name for each sequence in your order. You will also be able to specify different scales and Trityl OFF/ON requests for each oligo. See the screen capture below:

In the above example, a 5' FAM is requested. Use both the "Sequence" and "Modification Information" fields to indicate unambiguously what you want us to make!

In the next example, an oligo with two internal modifications is requested. Here is a good way to accomplish this - See the screen capture below:

In the above example, characters "X" and "Y" are used within the sequence to identify particular modifications. A key to what modifications these characters represent is given in the Modification Information field. Be sure not to use the same characters to indicate different modifications within your order!

When ordering chimeric oligos containing both RNA and DNA, it is best to use lowercase for DNA and UPPERCASE for RNA. Then indicate "lowercase=DNA, UPPERCASE=RNA" in the Modification Information field.

When indicating a phosphorothioate linkage, please use an "*" BETWEEN each base to show where the linkage should occur. This is shown in the following example:

The above examples are all good ways to descibe what type of modification you want. However, feel free to use the "Sequence" and "Modification Information" fields any way you wish, but please be sure that each modification and its position is clear.

Once you have filled out all of your oligo information, click "Place Modified Oligo Order" at the bottom of the form.
