Large Scale (0.5 mmol) Peptide Synthesis

(Please note: all prices listed are for Yale investigators only. If you are submitting a sample or order from outside of Yale, please check the price list for appropriate charges.)

Synthetic peptides can be made routinely up to 40 residues and often, depending on sequence, up to 70 residues. All peptides that can be purified are chromatographed on a preparative C-18 or C-4 RP-HPLC system and delivered as a lyophilized material. Yields for normal peptides under 40 residues are "guaranteed" at 50 mg or more and at 90+% purity. Yields and purity are often higher, varying with the peptide sequence and length. For peptides over 40 residues or for peptides with unusual sequences, the "guaranteed" yields and purity will be considered on an individual basis.

All peptides made in the facility at the 0.5 mmole scale are done by tBOC chemistry and at a single scale range (which generates a theoretical crude yield of 500-1,000 mg for a 10-20 mer respectively). For the incorporation of unusual amino acids, such as non-radioactive isotopes, the appropriate tBOC and side-chain protected material must be supplied by the submitter at a level of 2 mmoles/residue.

The turn-around time is 4 weeks/peptide/laboratory (P.I.), on the average. Thus, a request for 3 peptides from a single laboratory could take as long as 12 weeks if there is a large backlog. The purified peptides are characterized by analytical RP-HPLC, amino acid analysis, and laser desorption or FAB mass spectroscopy. The following service charges apply to Yale University investigators (see complete list of fees for surcharge prices for non-Yale requests):   [Order Forms]

              Residues 1 - 20 $ 55/residue
              Residues 21 - 50 83/residue
              Residues 51 + 110/residue
              Acetylation 55
              Cys, His, Trp 55 extra/residue
              Resin splitting 110/split
              Cleavage/characterization* 495/peptide
*This is a "package" which is required for each peptide that is requested whether the peptide is 2 residues or 70 residues. It includes the cleavage, purification, and all characterizations. For example, a 25 residue peptide with no Cys, His or Trp, would cost (20 x 55) + (5 x 83) + (1 x 495) = $2010.
